QTL analysis and development of HRM markers associated with fruit shape in interspecific pears (Pyrus pyrifolia × P. bretschneideri)

چکیده

Fruit shape is one of the important quantitative traits in pear (Pyrus spp.) breeding program, thus genetic study related to fruit could be beneficial breeding. Quantitative trait loci (QTL) analysis was carried out using ‘Whangkeumbae’ (P. pyrifolia, round) × ‘Yali’ bretschneideri, pyriform) and high-resolution melting (HRM) markers were developed. The linkage map constructed single nucleotide polymorphisms (SNPs) derived from Axiom Pear 70 K Genotyping Array simple sequence repeats. integrated showed ~ 90% genome coverage, a total distance 998.2 cM, marker density 1.6 cM. F1 progenies normal distribution length (L), diameter (D), L/D ratio. Three QTLs located group (LG) 6, 7, 12 identified with LOD thresholds 2.8–3.0. Six HRM developed array-SNPs anchored predicted 28.6–65.3% accuracy. Notably, accuracy increased by an combination consisting CBp06sn02, CBp07sn01, CBp12sn03. These results provide better understanding mechanism development reducing period.

Development, characterization, and annotation of potential simple sequence repeats by transcriptome sequencing in pears (Pyrus pyrifolia Nakai).

Simple sequence repeats (SSRs), one of the most powerful molecular markers, can be used for DNA fingerprinting, variety identification, genetic mapping, and marker-assisted selection. Using the pear's (Pyrus pyrifolia Nakai) 75,764 unigenes (55,676,271 bp) obtained by deep transcriptome sequencing, a total of 10,622 novel SSRs were identified in 9154 unigenes, accounting for 14.02% of all unige...

Identification of QTLs controlling harvest time and fruit skin color in Japanese pear (Pyrus pyrifolia Nakai)

Using an F1 population from a cross between Japanese pear (Pyrus pyrifolia Nakai) cultivars 'Akiakari' and 'Taihaku', we performed quantitative trait locus (QTL) analysis of seven fruit traits (harvest time, fruit skin color, flesh firmness, fruit weight, acid content, total soluble solids content, and preharvest fruit drop). The constructed simple sequence repeat-based genetic linkage map of '...

MYB Transcription Factors in Chinese Pear (Pyrus bretschneideri Rehd.): Genome-Wide Identification, Classification, and Expression Profiling during Fruit Development

The MYB family is one of the largest families of transcription factors in plants. Although, some MYBs were reported to play roles in secondary metabolism, no comprehensive study of the MYB family in Chinese pear (Pyrus bretschneideri Rehd.) has been reported. In the present study, we performed genome-wide analysis of MYB genes in Chinese pear, designated as PbMYBs, including analyses of their p...

Pollen density on the stigma affects endogenous gibberellin metabolism, seed and fruit set, and fruit quality in Pyrus pyrifolia

To clarify the relationship between pollen density and gametophytic competition in Pyrus pyrifolia, gametophytic performance, gibberellin metabolism, fruit set, and fruit quality were investigated by modifying P. pyrifolia pollen grain number and density with Lycopodium spores. Higher levels of pollen density improved seed viability, fruit set, and fruit quality. Treatments with the highest pol...

De novo assembly, functional annotation, and marker development of Asian pear (Pyrus pyrifolia) fruit transcriptome through massively parallel sequencing.

This study investigated the Asian pear transcriptome using the RNA-Seq normalized fruit cDNA library to create a transcriptomic resource for unigene and marker discovery. Following the removal of lowquality reads, 127,085,054 trimmed reads were assembled de novo to yield 37,649 non-redundant unigenes with an average length of 599 bp. Alternative splicing events were detected in 4121 contigs. A ...
